Glycosylation is a common posttranslational modification of proteins where glycan chains are added to a protein molecule. More than 50% of proteins are glycosylated resulting in changes in the protein such as folding, targeting, ligand recognition, binding, stability, immunogenicity, biological activity, etc.

Today, glycans play a significant biological role in four major medical areas; cancer, immunological disease, infectious disease and regenerative medicine. Glycoproteins show promise in the area of cancer biomarkers. Approximately 50% of current cancer markers widely used in clinical cancer diagnosis, are, in fact, various types of Glyco-markers. It is common knowledge in the infectious disease community that most communicable viruses target receptor glycoproteins formed on cell membranes. This fact provides drug developers with opportunities to develop vaccines directed at these target glycoproteins. These are only a few of the many examples that illustrate the need for a quick, simple and highly sensitive method for profiling and analyzing complex glycan structures which can then be used by the biopharmaceutical industry to accelerate the development of advanced biomarkers, thereapeutic proteins and stem cell characterization methodologies; all leading to improved diagnostics and therapeutic treatments.

Based on this market need, Moritex developed a novel, high throughput and highly sensitive glycan profiling instrument based on the principle of evanescent-field fluorescence excitation in combination with lectin microarrays.

GlycoStation™ Reader 1200, Evanescent-field Fluorescence Excitation Glycan Profiler

GlycoStation™ Reader 1200GlycoStation™ Reader 1200 is an ultra-high speed and highly sensitive optical microarray scanner utilizing the principle of evanescent-field fluorescence excitation to scan an entire microarray slide in only two minutes. GlycoStation™ Reader 1200 is designed to interface with a novel glycan profiling system that evaluates even weak molecular interactions from a wash free lectin microarray thereby reducing the total time required to scan, profile and analyze a biological sample.

LecChip™ Ver. 1.0 Lectin Microarrays

LecChip™ Ver. 1.0 Lectin MicroarraysThe LecChip™ Ver 1.0 lectin microarrays contain 45 lectins, carefully selected from a candidate pool of 167 lectins. Lectins are well known as glycan recognizers and are classified into several categories, for instance; fucose, sialic acid, asialo-form, agalacto-form, high mannose, O-glycan and branching structure recognizers among others.

LecChip™ Ver 1.0 microarrays feature 7 wells on a glass slide with 45 lectins in each well. Each lectin is spotted 3 times resulting in a total of 135 spots, excluding position markers, in each well (45 lectins × 3 spots = 135 spots)

Differential Glycan Profiling Analysis Software:GlycoStation™ Tools Ver.1.0

GlycoStation™ Tools Ver.1.0 is a graphical display software used to differentially compare two or more samples. Bar graphs and binding curves for each lectin, as well as data export for other applications are standard features.

In order to maximize the S/N ratios of the glycan profiling patterns, Moritex highly recommends the use of Moritex’s proprietary probing solution for the lectin microarrays.
